DESCRIPTION

Lorabid® (loracarbef, USP) is a synthetic?-lactam antibiotic of the carbacephem class for oral administration. Chemically, carbacephems differ from cephalosporin-class antibiotics in the dihydrothiazine ring where a methylene group has been substituted for a sulfur atom.

The chemical name for loracarbef is: (6R, 7S)-7-[(R)-2-amino-2-phenylacetamido]-3-chloro-8-oxo-1-azabicyclo[4.2.0]oct-2-ene-2-carboxylic acid, monohydrate. It is a white to off-white solid with a molecular weight of 367.8. The empirical formula is C16H16ClN3O4•H2O. The structural formula is:

Lorabid Pulvules® (loracarbef capsules, USP) and Lorabid for Oral Suspension (loracarbef for oral suspension, USP) are intended for oral administration only.

Each Pulvule contains loracarbef equivalent to 200 mg (0.57 mmol) or 400 mg (1.14 mmol) anhydrous loracarbef activity. They also contain cornstarch, dimethicone, FD & C Blue No. 2, gelatin, iron oxides, magnesium stearate, and titanium dioxide.

What is leucovorin?

Leucovorin is a form of folic acid. Folic acid helps your body produce and maintain new cells, and also helps prevent changes to DNA that may lead to cancer. Folic acid is present in many foods such as dried beans, peas, oranges, spinach, and others.

Leucovorin is used to reduce the side effects of large doses or accidental overdose of medications that can reduce the effects of folic acid in the body. This includes methotrexate (Rheumatrex), pyrimethamine (Daraprim), trimethoprim (Trimpex, Proloprim), and others.

Lubiprostone is used for:

Treating chronic constipation. It is also used to treat irritable bowel syndrome with constipation (IBS-C) in women.It may also be used for other conditions as determined by your doctor.

Lubiprostone is a chloride channel activator. It works by increasing fluid secretion in the intestine, which increases intestinal muscle movement and helps you to pass the stool.

Do NOT use Lubiprostone if:

  • you are allergic to any ingredient in Lubiprostone
  • you have severe diarrhea or stomach or bowel blockage

Before using Lubiprostone:

Some medical conditions may interact with Lubiprostone. Tell your doctor or pharmacist if you have any medical conditions, especially if any of the following apply to you:

  • if you are pregnant, planning to become pregnant, or are breast-feeding
  • if you are taking any prescription or nonprescription medicine, herbal preparation, or dietary supplement
  • if you have allergies to medicines, foods, or other substances
  • if you have diarrhea or a history of stomach or bowel blockage
  • if you have liver or kidney problems

Linezolid is used for:

Treating adults with certain serious bacterial infections that are often resistant to other antibiotics.

Linezolid is an oxazolidinone antibiotic. It works by interfering with the production of proteins needed by bacteria to grow.

Do NOT use Linezolid if:

  • you are allergic to any ingredient in Linezolid
  • you are taking an amphetamine (eg, dextroamphetamine), an anorexiant (eg, phentermine), a catechol-O-methyltransferase (COMT) inhibitor (eg, entacapone), cyclobenzaprine, fentanyl, levodopa, meperidine, sibutramine, a sympathomimetic (eg, albuterol, pseudoephedrine), tetrabenazine, or a triptan (eg, sumatriptan)
  • you are taking or have taken apraclonidine, bupropion, buspirone, dextromethorphan, a monoamine oxidase inhibitor (MAOI) (eg, phenelzine, selegiline), nefazodone, a norepinephrine reuptake inhibitor (eg, atomoxetine), a serotonin-norepinephrine reuptake inhibitor (SNRI) (eg, duloxetine, venlafaxine), a selective serotonin reuptake inhibitor (SSRI) (eg, paroxetine), a tetracyclic antidepressant (eg, maprotiline, mirtazapine), trazodone, a tricyclic antidepressant (eg, amitriptyline, doxepin), or vilazodone within the past 14 days
  • you are taking or have taken fluoxetine within the past 5 weeks

Letairis is very likely to produce serious birth defects if used by pregnant women, as this effect has been seen consistently when it is administered to animals [see Contraindications (4.1)]. Pregnancy must therefore be excluded before the initiation of treatment with Letairis and prevented during treatment and for one month after stopping treatment by the use of two acceptable methods of contraception unless the patient has had a tubal sterilization or chooses to use a Copper T 380A IUD or LNg 20 IUS, in which case no additional contraception is needed. Obtain monthly pregnancy tests.

Because of the risk of birth defects, Letairis is available only through a special restricted distribution program called the Letairis Education and Access Program (LEAP), by calling 1-866-664-LEAP (5327). Only prescribers and pharmacies registered with LEAP may prescribe and distribute Letairis. In addition, Letairis may be dispensed only to patients who are enrolled in and meet all conditions of LEAP [see Warnings and Precautions (5.1)].

Indications and Usage for Letairis

Letairis is indicated for the treatment of pulmonary arterial hypertension (PAH) (WHO Group 1) to improve exercise ability and delay clinical worsening. Studies establishing effectiveness included predominantly patients with WHO Functional Class II–III symptoms and etiologies of idiopathic or heritable PAH (64%) or PAH associated with connective tissue diseases (32%).
